Product description

Full-length, Lambda Protein Phosphatase (Lambda PP) is the 221 amino-acid product from ORF221 of bacteriophage lambda. Lambda PP is a Mn2+-dependent protein phosphatase with activity towards phosphorylated serine, threonine, tyrosine and histidine residues. Lambda PP is expressed and purified from E. coli as a recombinant.

Applications

Lambda Protein Phosphatase can be used to release phosphate groups from phosphorylated serine, threonine and tyrosine residues in proteins.

Unit definition

One unit will hydrolyze 1 nmole of p-nitrophenyl phosphate per minute at 30 and pH 7.5.

50 mM HEPES, 100 mM NaCl, 2 mM DTT, 0.1 mM EDTA, 0.1 mM MnCl2, 0.01% Brij 35, 50% glycerol, pH 7.5

 

Cautions

- Repeated freeze/thaw cycles is not recommended.  

- For each individual substrate, optimal incubations times and enzyme concentrations must be established 

  experimentally in advance.

- In case of assays on crude samples, addition of protein inhibitor cocktail and shorter incubation time are 

  recommended.

- The phosphatase activity is inhibited at the high concentrations of sodium orthovanadate (> 50mM), sodium 

  fluoride (> 50 mM) and EDTA (> 50 mM).
