Description

EZ™ T7 In vitro mRNA Synthesis Kit of Enzynomics is a product making the process from transcription to 5’ capping and 3’ poly(A) tailing simple and efficient. It is a product specialized for the production of mRNA suitable for transfection and microinjection experiments. Containing Anti-Reverse Cap Analog (ARCA), it can synthesize 100% functional mRNA in contrast to 50% efficiency shown from mRNA synthesis with existing Cap analog. Because this product optimizes the ratio between ARCA and GTP in 4:1 via high-concentration NTP, it is possible to obtain a large amount of capped transcription product from one reaction. In addition, we increase convenience through providing RNA polymerase mixed with RNA inhibitor, which is for preventing RNA degradation caused by the contamination during transcription progress. 20~30 μg of mRNA can be produced per one reaction using 1 μg of Control Template, which is one of kit components. After Transcription & capping progress, 3’ poly(A)-tail reaction is executed by using E.coli Poly(A) Polymerase. During 30 minutes of reaction, more than 50-200 bp of Poly(A) can be ligated. With capped poly(A)-tailed RNA which is the final product of two-step experiment process, not only the stability of transfection and microinjection experiments but also translation efficiency can be increased.

Quality control

1 μg of linearized template was used in transcription and ARCA capping reaction.

Generated transcripts of 1 kb and 2 kb was poly(A)-tailed according to the protocol.

Synthesized mRNA was analyzed on 1% agarose gel with ethidium bromide.
