Product description

EZ™ ExoAP PCR Product Clean-up Mix utilizes two hydrolytic enzymes, Exonuclease I and Antartic Phosphatase (AP), together in a specially formulated buffer, to remove unwanted dNTPs and primers from PCR products. Exonuclease I removes residual single-stranded primers and any extraneous single-stranded DNA produced in the PCR. AP removes the remaining dNTPs from the PCR mixture. Kit reagent is designed for simple, quick PCR cleanup for downstream applications, such as DNA sequencing or Single Nucleotide Polymorphism (SNP) analysis. When PCR amplification is complete, any unconsumed dNTPs and primers remaining in the PCR product mixture will interfere with these methods. EZ™ ExoAP PCR Product Clean-up Mix removes these contaminants.

Applications

Kit reagent is designed for simple, quick PCR cleanup for downstream applications, such as DNA sequencing or Single Nucleotide Polymorphism (SNP) analysis. When PCR amplification is complete, any unconsumed dNTPs and primers remaining in the PCR product mixture will interfere with these methods.

(A) PCR clean-up enzyme (ExoAP from Enzynomics, Inc. or ExoSAP from Company A) treated or untreated 2 kb of PCR product was cloned using EZ-Fusion™ Cloning Kit (Cat# EZ015). Untreated control and vector only were used as a negative controls. (B) Success rate was measured by colony PCR.

1,860 bp fragment was amplified and treated with ExoAP reagent (above) or an alternate enzymatic reagent (below, ExoSAP) and sequenced. Pherograms demonstrated a miscall of sequence T to C with the alternate enzymatic reagent.
